Overview

Most of what is known about FoxO3 expression patterns comes from studies of Foxo3a in mice, although no comprehensive reports have been published. In developing mice, high levels of Foxo3a are found in the liver. Despite some contradictory results, in adult mice it is thought that Foxo3a is highly expressed in heart, brain, spleen and kidney tissues, as well as to a lesser extent in many other tissue and cell types including lung, liver, testis and ovary [1][2].

Foxo3a is also found within the brain, although during development Foxo3a is not found there until embryonic day 14.5 onwards [3]. It has a widespread expression throughout the brain (unlike Foxo1, for example, which is expressed only in specific areas), including in the hippocampus, the granule neurons of the cerebellum, and in the neocortex. It is possible that the distribution and expression levels of FOXO3 change over time even in adults, since the expression of FoxO3a in rat skeletal muscle has been shown to peak at 6 months of age [4].

In humans, FOXO3 is also thought to be distributed widely around the body, as it has been found in many cell types. However, expression (as well as activity) may be reduced in tumours [5].
