Overview

Foxh1 can directly interact with Smad2 and Smad3 to form DNA-binding complex in response to TGFβ/activin/nodal signals. Smad4 is also recruited to the complex although Smad4 is not thought to directly interact with Foxh1 but rather through Smad4’s interaction with the R-Smads. Drap1 has been shown to inhibit Foxh1 activity by binding to the forkhead domain and preventing DNA-binding. Thus in the early embryo, Drap1 functions to repress the activity of the nodal signalling by blocking Foxh1 functions [1]. Foxh1 also interacts with other transcription factors to both positively and negatively regulate transcription. For example, Foxh1 can form a complex with goosecoid (GSC) and HDAC to repress expression of Mixl1 [2]. Alternatively, Foxh1 can interact with Nkx2-5 to activate expression of the Mef2c gene [3]. The interactions with Gsc and Nkx2-5 are both mediated through the Foxh1 forkhead domain. Foxh1 can also interact with the forkhead factor, Foxg1 (also known as BF1) and prevent the interaction of Smads with Foxh1, thereby inhibiting signalling [4].
